Incidence and Molecular-Based Identification of Papaya ringspot virus Infecting Papaya in Java

Infeksi Papaya ringspot virus (PRSV) di Indonesia pertama kali dilaporkan pada pepaya Nangroe Aceh Darussalam tahun 2012. Sejak itu, PRSV atau penyakit bercak bercincin menyebar ke beberapa daerah Jawa, Sumatera, Bali, dan Nusa Tenggara Barat. Penelitian ini dilakukan untuk mengonfirmasi keberadaan penanaman Jawa mengetahui identitas molekulernya. Metode double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) dengan antiserum spesifik digunakan mendeteksi sampel dari lapangan. Insidensi hasil deteksi DAS-ELISA Bogor, Purworejo, Kebumen, Bantul berturut-turut sebesar 59.1%, 51.4%, 84.2%, 96.2%. Identifikasi lebih lanjut reverse transcription-polymerase chain reaction menggunakan primer (PRSV326/PRSV800), dilanjutkan sikuensing DNA. Fragmen DNA berukuran 475 pb berhasil diamplifikasi lapangan analisis nukleotida menunjukkan bahwa semua terkonfirmasi homologi antarisolat berkisar antara 95.4% sampai 99.4%. Analisis filogenetika isolat-isolat berada dalam satu grup yang sama isolat PRSV-P Thailand.

The complete genome sequence (10,326 nucleotides) of a papaya ringspot virus isolate infecting genetically modified papaya in Hainan Island of China was determined through reverse transcription (RT)-PCR. The virus shares 92% nucleotide sequence identity with the isolate that is unable to infect PRSV-resistant transgenic papaya.
